deamidation raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2026-04-04. Anything still debated is marked as such rather than presented as settled.
Common degradation routes include hydrolysis of labile amide bonds, deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation. Aggregates can form during freeze-thaw cycling, at elevated pH, or when peptide concentration is high. Each route produces characteristic chromatographic or mass shifts that are tracked during stability studies. Whether a given minor impurity alters biological activity is often an open question, and specification limits are typically set on identity and purity rather than on functional data for trace species.
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.

Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.
Long-term storage of lyophilised peptide powder is generally at minus twenty degrees Celsius or colder, with desiccant and protection from light. Short-term storage at two to eight degrees Celsius is common during active use. In solution, stability depends strongly on pH, concentration, and the presence of preservatives, and hydrolysis or aggregation can develop over weeks. Published stability data specific to this molecule are limited, so recommended conditions for research material are usually extrapolated from general peptide handling practice rather than from a dedicated study.
Bulk peptide material is normally characterised by reversed-phase high-performance liquid chromatography, which separates the target sequence from truncation products and other closely related impurities. Ultraviolet detection near 214 nanometres is common because the peptide backbone absorbs in that region. Mass spectrometry, usually electrospray ionisation coupled to a mass analyser, is used to confirm the molecular mass. Because the molecule carries a lipophilic side chain, gradient methods often need a relatively high organic modifier fraction to elute it within a practical retention window.
== Treatment == Traditionally, nothing by mouth was considered to be mandatory in all cases, but gentle feeding by enteral feeding tube may help to restore motility by triggering the gut's normal feedback signals, so this is the recommended management initially. When the patient has severe, persistent signs that motility is completely disrupted, nasogastric suction and parenteral nutrition may be required until passage is restored. In such cases, continuing aggressive enteral feeding causes a risk of perforating the gut. Several options are available in the case of paralytic ileus. Most treatment is supportive. If caused by medication, the offending agent is discontinued or reduced. Bowel movements may be stimulated by prescribing lactulose, erythromycin or, in severe cases that are thought to have a neurological component (such as Ogilvie's syndrome), neostigmine. There is also evidence from a systematic review of randomized controlled trials that chewing gum, as a form of 'sham feeding', may stimulate gastrointestinal motility in the post-operative period and reduce the duration of postoperative ileus. If possible, the underlying cause is corrected (e.g. replace electrolytes).
Fumaric acid or trans-butenedioic acid is an organic compound with the formula HO2CCH=CHCO2H. A white solid, fumaric acid occurs widely in nature. It has a fruit-like taste and has been used as a food additive. Its E number is E297. The salts and esters are known as fumarates. Fumarate can also refer to the C4H2O2−4 ion (in solution). Fumaric acid is the trans isomer of butenedioic acid, while maleic acid is the cis isomer.

=== Leo Bloom === Leo Bloom (Sonny Poon Tip) is Jesse's wayward, estranged 19-year-old son. Jesse hires Gus to tutor Leo on his college admissions essays to Oxford and Cambridge despite Leo's disinterest in school, and Leo and Gus soon begin a sexual relationship. Over time, Gus helps Leo discover his academic ambitions, and Leo is grateful for the experience despite not being admitted to either of his choice schools. Gus later utilizes his connections at Oxford to land Leo an admission, and Jesse hires Gus as his assistant in return after Gus loses his government job.
== History == Throughout most of human history, infant nutrition has primarily depended on the availability of the child's mother or a wet nurse to breastfeed the infant. Beliefs and behaviors relating to infant feeding also vary widely across countries, cultures and times. Mothers and caregivers have also sought additional ways to feed children, sometimes referred to as "hand feeding". As early as 1500 BCE, Egyptian pottery shows images of women breastfeeding their babies and also using animal horns to feed them.
== Medical use and research == Epothilone D, with the generic drug name utidelone, was approved in China in 2021 for the treatment of metastatic breast cancer. Utidelone has shown benefits in a phase III breast cancer trial when added to capecitabine. One synthetic analog, ixabepilone, was approved in October 2007 by the United States Food and Drug Administration for use in the treatment of aggressive metastatic or locally advanced breast cancer that no longer responds to currently available chemotherapies. In November 2008, the EMEA refused a marketing authorization for ixabepilone. Epothilone B, with the generic drug name patupilone, was proven to contain potent in vivo anticancer activities at tolerated dose levels in several human xenograft models. As a result, patupilone and various analogs underwent various clinical phases. Patupilone and the fully synthetic sagopilone were tested in phase II trials and BMS-310705 was tested in phase I trials). Patupilone failed a phase III trial for ovarian cancer in 2010. Results of a phase III trial with ixabepilone (BMS-247550) in combination with capecitabine in metastatic breast cancer have been announced (2007 – leading to FDA approval).

==== The phenethylamine psychedelic mescaline ==== Mescaline is sometimes described as the "first psychedelic", as it was the first to be discovered and characterized by the Western world. American physician John Raleigh Briggs, living in Texas, learned of peyote from Native Americans and Mexicans, who told him that it produced "beautiful visions" and made them journey into the "spirit world". He obtained mescal buttons from Mexico and published a journal article about trying a very low dose of them in May 1887. This article is said to have brought peyote into North American pharmacology. Briggs described the physiological symptoms of his experience, such as dramatically increased heart rate and of experiencing "intoxication". However, a modern interpretation is that Briggs simply had a severe panic attack in response to the drug. In any case, his article was read by George Davis, of Parke, Davis and Company, who then obtained the buttons from Briggs in June 1887. Parke-Davis attempted to market peyote as a cardiac stimulant and for other uses, but met with little success. The German pharmacologist Louis Lewin obtained mescal buttons from Parke-Davis during a trip to the United States in 1887 and began studying them and sharing his findings. James Mooney, the first Westerner known to have participated in a Native American peyote ceremony in 1892, distributed peyote to a number of authorities in 1894. Using these specimens, the hallucinogenic effects of peyote were first properly described by Daniel W. Prentiss and F. P. Morgan in 1895.
=== Post-AFL career === In 2013, Prismall joined the Western Bulldogs. On-field, he played in 2013 for the Bulldogs' VFL affiliate, the Williamstown Seagulls; then, in 2014, when the Bulldogs ended their affiliation with Williamstown, began playing for the club's new reserves team in the VFL. He served in other roles at the football club, including as a runner and as player welfare manager. Prismall, along with 33 other Essendon players, was found guilty of using a banned performance-enhancing substance, thymosin beta-4, as part of Essendon's sports supplements program during the 2012 season. He and his teammates were initially found not guilty in March 2015 by the AFL Anti-Doping Tribunal, but a guilty verdict was returned in January 2016 after an appeal by the World Anti-Doping Agency. He was suspended for two years which, with backdating, ended in November 2016; as a result, he served approximately fourteen months of his suspension and missed the entire 2016 VFL season and was forced to stand down from his role as player welfare manager. After 13 years working in player welfare at the Bulldogs, Prismall returned to Essendon ahead of the 2027 AFL season as the club's Head of Development.
The Juárez Cartel controls one of the primary transportation routes for billions of dollars' worth of illegal drug shipments annually entering the United States from Mexico. Since 2007, the Juárez Cartel has been locked in a vicious battle with its former partner, the Sinaloa Cartel, for control of Ciudad Juárez. La Línea is a group of Mexican drug traffickers and corrupt Juárez and Chihuahua state police officers who work as the armed wing of the Juárez Cartel. Vicente Carrillo Fuentes headed the Juárez Cartel until his arrest in 2014. Since 2011, the Juárez Cartel has continued to weaken. It is present in the three main points of entry into El Paso, Texas. Its weakness and inability to effectively fight against Sinaloa's advances in Juárez contributed to the lower death toll in Juárez in 2011.
=== Cell design === Operando spectroscopy requires measurement of the catalyst under (ideally) real working conditions, involving comparable temperature and pressure environments to those of industrially catalyzed reactions, but with a spectroscopic device inserted into the reaction vessel. The parameters of the reaction are then measured continuously during the reaction using the appropriate instrumentation, i.e., online mass spectrometry, gas chromatography or IR/NMR spectroscopy. Operando instruments (in situ cells) must ideally allow for spectroscopic measurement under optimal reaction conditions. Most industrial catalysis reactions require excessive pressure and temperature conditions which subsequently degrades the quality of the spectra by lowering the resolution of signals. Currently many complications of this technique arise due to the reaction parameters and the cell design. The catalyst may interact with the components of the operando apparatus; open space in the cell can have an effect on the absorption spectra, and the presence of spectator species in the reaction may complicate analysis of the spectra. Continuing development of operando reaction-cell design is in line with working towards minimizing the need for compromise between optimal catalysis conditions and spectroscopy. These reactors must handle specific temperature and pressure requirements while still providing access for spectrometry.
Microbial growth and proliferation can be inhibited by a technique called biopreservation. Biopreservation is achieved by adding antimicrobials or by increasing the acidity of the fish muscle. Most bacteria stop multiplying when the pH is less than 4.5. Acidity is increased by fermentation, marination or by directly adding acids (acetic, citric, lactic) to fish products. Lactic acid bacteria produce the antimicrobial nisin which further enhances preservation. Other preservatives include nitrites, sulfites, sorbates, benzoates and essential oils.

Reversed-phase high-performance liquid chromatography is the standard approach, separating the main peak from related impurities. Ultraviolet detection near 214 nanometers captures the peptide backbone. Mass spectrometry is then used alongside chromatography to confirm identity and detect covalent modifications.
Cycling between frozen and liquid states concentrates the peptide at ice interfaces, which favors non-covalent association. The resulting aggregates may be invisible to simple assays yet alter recovery and apparent potency. Limiting the number of cycles and aliquoting before storage are common mitigations.
Tryptophan and methionine side chains can undergo photo-induced oxidation, so amber glass or opaque packaging is typical. The effect is gradual and depends on wavelength and exposure time. Light protection is usually specified for both solid and solution forms.
Reversed-phase liquid chromatography with ultraviolet detection is the usual approach, frequently combined with mass spectrometry for identity. Purity is reported as the area percentage of the main peak. Related impurities eluting near the main peak are usually summed and reported separately.
