Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Long-term storage of the solid generally relies on temperatures at or below minus twenty degrees Celsius, while short-term working stocks may be held refrigerated. Light exposure is limited because photodegradation can alter side chains over extended periods. Solutions prepared for analysis are less stable than the dry powder and are typically used within the same working day. Buffer choice matters, since some aqueous conditions favor deamidation or oxidation at specific residues. Stability data are usually generated under defined accelerated conditions and then extrapolated with stated assumptions.
Identity and purity are established with reversed-phase high-performance liquid chromatography, often paired with mass spectrometry for confirmation of the expected mass. Peptide mapping after enzymatic digestion verifies the primary sequence and detects substitutions. Size-exclusion chromatography quantifies aggregates and fragments, which are the impurities most often tracked for peptides of this size. Residual solvents, counterions, and water content fall under separate tests described in pharmacopeial chapters. Circular dichroism or nuclear magnetic resonance may be used in research settings to probe secondary structure, though such methods are less common in routine release testing.

Clinical research programs have evaluated tirzepatide in adults with type 2 diabetes and in adults with obesity or excess weight. Trials generally reported reductions in glycated hemoglobin and body weight across treatment periods of several months. Since these studies enrolled defined populations under controlled conditions, the findings describe group averages rather than individual outcomes. Open questions include the durability of effects after treatment stops, variation among subgroups, and the long-term consequences of sustained dual receptor stimulation. Published trial summaries should be consulted for exact measurements rather than secondary accounts.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its backbone derives from the native glucose-dependent insulinotropic polypeptide sequence, altered at several positions to resist enzymatic cleavage. A fatty diacid group attached through a linker extends plasma residence time by promoting reversible binding to serum albumin. The molecule carries a net negative charge near physiological pH and has a reported molecular weight close to 4813 daltons. These features separate it from shorter incretin analogs and account for its prolonged dosing interval.
Pharmacologically, tirzepatide activates two distinct G protein-coupled receptors: the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Binding at each target triggers cyclic AMP accumulation and downstream signaling in pancreatic beta cells, adipose tissue and the central nervous system. Because the two pathways overlap only partially, the combined effect on insulin secretion, glucagon suppression and appetite signaling differs from that of selective single-receptor compounds. Affinity is not equal across the two targets, and the clinical meaning of that imbalance remains an area of active study.
Both receptors are class B G protein-coupled receptors that signal largely through Gs-mediated cyclic AMP production. Activation within pancreatic islets increases glucose-dependent insulin secretion and suppresses glucagon release when glucose is elevated. Outside the pancreas, signaling in the central nervous system and gut appears to influence appetite and gastric emptying. The relative contribution of each receptor to observed clinical effects remains under investigation, and the two pathways are not simply additive in practice.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.
=== Ultimate Fighting Championship === On July 18, 2015, Casey debuted in the UFC to face The Ultimate Fighter: A Champion Will Be Crowned contestant Joanne Calderwood, at UFC Fight Night: Bisping vs. Leites, replacing the injured Bec Rawlings. After winning round one, Casey was outpointed by Calderwood in the next two rounds and lost a unanimous decision. Despite the loss, she was awarded a "Fight of the Night" bonus award. Casey next faced Seo Hee Ham at UFC Fight Night: Henderson vs. Masvidal on November 28, 2015. Again Casey lost a unanimous decision, but was awarded "Fight of the Night". Casey faced Cristina Stanciu at UFC Fight Night: McDonald vs. Lineker on July 13, 2016. She earned her first UFC win by defeating Stanciu by TKO in the first round. Casey next faced Randa Markos at UFC 202 on August 20, 2016. Casey caught Markos in an armbar, to which she verbally submitted, giving Casey back-to-back UFC wins. Casey next faced Cláudia Gadelha at UFC Fight Night: Bader vs. Nogueira 2. She lost the fight by unanimous decision. During the third round there was controversy when Gadelha appeared to land an illegal head kick on Casey while Casey was grounded. However, the referee deemed the kick unintentional and no point was deducted. A replay showed that Gadelha's shin appeared not to hit Casey's skull but caught her hair instead. Casey was criticized for allegedly trying to play up the illegal head kick and possibly achieve a disqualification win. Casey next faced Jessica Aguilar at UFC 211 on May 13, 2017. She won the fight via unanimous decision.

Polyendocrine metabolic ovarian syndrome (PMOS) is an endocrine disorder characterized by an excess of androgens produced by the ovaries. It is estimated that approximately 90% of women with PMOS demonstrate hypersecretion of these hormones. The cause of this condition is unknown. Speculations include genetic predisposition; however, the gene or genes responsible for this remain unidentified. The condition may have a hereditary basis. Other possible causes include elevated insulin production. Most cases of PMOS involve insulin resistance. It is thought that adipose tissue dysfunction plays a role in the insulin resistance seen in PMOS. Insulin can induce excess testosterone secretion from the ovaries. A complication associated with polyendocrine metabolic ovarian syndrome is high cholesterol, which is treated with statins. In a meta-analysis, atorvastatin was shown to decrease androgen concentrations in people with hyperandrogenism. Elevated insulin leads to lower production of sex hormone binding globulin (SHBG), a regulatory glycoprotein that suppresses the function of androgens. High blood levels of insulin also work in conjunction with ovarian sensitivity to insulin to cause hyperandrogenemia, the primary symptom of PMOS. Obese individuals may be more biologically inclined to PMOS due to markedly higher insulin. This hormonal imbalance can lead to chronic anovulation, in which the ovaries fail to release mature eggs. These cases of ovulatory dysfunction are linked to infertility and menstrual disturbances.

== Medical uses == Alfatradiol is used in form of an ethanolic solution for topical application on the scalp. Similarly to other drugs against alopecia, topical or oral, it has to be applied continuously to prevent further hair loss. Regrowth of hair that was already lost is only possible to a limited extent. In general, advanced alopecia does not respond well to medical treatment, which has historically been thought to be a consequence of the hair roots being lost. A university-led study (including several authors who are advisors to companies such as Pfizer) in 103 women comparing alfatradiol to minoxidil, another topical hair loss treatment, found the latter to be more effective. In contrast to minoxidil, alfatradiol did not result in an increase of hair density or thickness, but only in slowing down or stabilization of hair loss in this study. In an earlier study, no systemic side effects were noted, and 17α-estradiol was found to reduce androgenic hair loss, though it was not effective at growing new hair. Other efforts of alfatradiol had been directed at neurodegenerative diseases including Parkinson's. Other hair loss medications include ketoconazole, finasteride, and dutasteride.
In spite of these warnings, the Luftwaffe did little to address this and treated the entire topic with some level of disdain. Their own radars were superior to CH in many ways, but had proven to be only marginally useful. During the Air Battle of the Heligoland Bight in 1939, a German Freya radar detected the raid while it was still an hour away from its target, yet had no way to report this to any of the fighter units that could intercept it. Getting the information from the radar to the pilots in a useful form appeared to be a difficult problem, and the Germans believed the British would have the same difficulty and thus radar would have little real effect. Some desultory effort was put into attacking the CH stations, especially during the opening stages of the Battle. British engineers were able to quickly return these units to service, or in some cases simply pretend to do so in order to fool the Germans into thinking the attacks failed. As the pattern of these attacks became clear, the RAF began to counter them with increasing effectiveness. The Junkers Ju 87 dive bombers were subjected to catastrophic losses and had to be withdrawn from battle. The Germans gave up trying to attack CH directly on any reasonable scale. Thus, CH was allowed to operate throughout the Battle largely unhindered. Although communications were indeed a serious problem, it was precisely this problem that the Dowding system had been set up to address, at great expense. The result was that every British fighter was roughly twice or perhaps more as effective than its German counterpart.
=== French === Union Saint-Jean-Baptiste - Founded March 27, 1900 in Woonsocket, Rhode Island, as the Union Saint-Jean-Baptiste d'Amerique for Roman Catholic Franco-Americans. Activities include hospital volunteer work, comforting the bereaved, visiting shut-ins, and working in Catholic Action. The Saint-Jean-Baptiste Educational Foundation provides scholarships; the group also helps fund the Catholic Communications Foundation whose mission was to spread understanding of the Catholic faith and doctrine in the mass media. Headquarters was Woonsocket. The motto is "In Union there is strength". Lodges are called local councils, and the national convention is "National Congress". There was a ritual for initiation and the installation of officers. 62,000 members in 1968, 47,000 in January 1979. Merged with Catholic Family Life in 1991. Association Canado-Americaine - Founded in 1896 in Manchester, New Hampshire, which remained the organization's headquarters. Lodges were called Courts, regions District Courts, highest body "Supreme Court" which met quadrennially. A "High Court" administered the group in between sessions of the Supreme Court and determined district boundaries. Motto: "Religion, Patriotism, and Fraternity." The Association had rituals for initiation, installation, and other rites; the rituals reflected the Catholic values of the society whose patron was St. John the Baptist. The organization offered beneficiary and social membership; the former consisted of adult and infant divisions, the infants becoming adult members when they turned 18.
==== C. Nasal blood supply – arteries and veins ==== Like the face, the human nose is well vascularized with arteries and veins, and thus supplied with abundant blood. The principal arterial blood-vessel supply to the nose is two-fold: (i) branches from the internal carotid artery, the branch of the anterior ethmoidal artery, the branch of the posterior ethmoidal artery, which derive from the ophthalmic artery; (ii) branches from the external carotid artery, the sphenopalatine artery, the greater palatine artery, the superior labial artery, and the angular artery. The external nose is supplied with blood by the facial artery, which becomes the angular artery that courses over the superomedial aspect of the nose. The sellar region (sella turcica, "Turkish chair") and the dorsal region of the nose are supplied with blood by branches of the internal maxillary artery (infraorbital artery) and the ophthalmic arteries that derive from the internal common carotid artery system. Internally, the lateral nasal wall is supplied with blood by the sphenopalatine artery (from behind and below) and by the anterior ethmoid artery and the posterior ethmoid artery (from above and behind). The nasal septum also is supplied with blood by the sphenopalatine artery, and by the anterior and posterior ethmoid arteries, with the additional circulatory contributions of the superior labial artery and of the greater palatine artery.
Jennings was also appointed as a pitchman for Wisconsin even outside the specialized burger promotion, in a contract signed on October 1, 2009; in exchange, A&W donated $10,000 to The Greg Jennings Foundation. In January 2013, A&W Restaurants appointed Lexington-based advertising agency Cornett Integrated Marketing as its new agency of record. The contract was renewed in January 2024. In January 2019, ahead of its hundredth anniversary, A&W removed the ampersand from its logo as part of a tongue-in-cheek campaign to bring back the ampersand to the alphabet as its 27th letter. Beginning in October 2020, in order to recoup economic losses from the closure of several of its restaurants, A&W set up a series of quizzes on its website. For Halloween that year, it released a web stunt called The Onion Ring, a parody of The Ring. A&W unveiled a new campaign in 2024, capitalizing on the ampersand of its logo and the tagline Burgers, Floats & Then Some. Packaging was also changed accordingly.

Between the 1960s and the 2010s, the percentage of children living with their married parents in their first marriage declined from approximately three quarters to just under half. At the same time, the share of children raised by single parents rose sharply. During the 2000s and 2010s, the number of American children raised by their grandparents or other family members rather than their parents also grew. American parents of the early twenty-first century usually do not insist that their children share the same political or religious beliefs, to follow the same traditions, or to get married and have children. Rather, they emphasize ethical behavior, tolerance, generosity, and financial independence. Parents from wealthier backgrounds are less likely to have children out of wedlock and more likely to stay married, with desirable outcomes for their children, including better social and cognitive development and educational attainment. By contrast, children born into the middle class or the lower class are less likely to have married parents than before. They also tend to have younger parents who did not intend to have them and more siblings. Upper middle-class and wealthy American couples living in the cities tend to have fewer children and to invest heavily in their children in the form of breastfeeding for at least a year, giving them premium healthcare plans, sending them to private schools, and letting them eat organic foods.

Phthalimide is the organic compound with the formula C6H4(CO)2NH. It is the imide derivative of phthalic anhydride. It is a sublimable white solid that is slightly soluble in water but more so upon addition of base. It is used as a precursor to other organic compounds as a masked source of ammonia.
=== Other indications === Sertraline is approved for the treatment of post-traumatic stress disorder (PTSD). The UK's National Institute for Clinical Excellence recommends it for patients who prefer drug treatment to a psychological one. Other guidelines also suggest sertraline as a first-line option for pharmacological therapy. When necessary, long-term pharmacotherapy can be beneficial. There are both negative and positive clinical trial results for sertraline, which may be explained by the types of psychological traumas, symptoms, and comorbidities included in the various studies. Positive results were obtained in trials that included predominantly women (75%) with a majority (60%) having physical or sexual assault as the traumatic event. Somewhat contrary to the above suggestions, a meta-analysis of sertraline clinical trials for PTSD found it to be statistically superior to placebo in the reduction of PTSD symptoms but the effect size was small. Another meta-analysis relegated sertraline to the second line, proposing trauma focused psychotherapy as a first-line intervention. The authors noted that Pfizer had declined to submit the results of a negative trial for inclusion in the meta-analysis, making the results unreliable. Sertraline, when taken daily, can be useful for the treatment of premature ejaculation. A disadvantage of sertraline is that it requires continuous daily treatment to delay ejaculation significantly.

It is a synthetic 39-amino-acid peptide that acts on two incretin receptors, the GIP receptor and the GLP-1 receptor. It is given by subcutaneous injection and has a circulating half-life of roughly five days. It is not a small molecule and is not absorbed usefully from the gut in conventional oral form.
Selective agents act on one receptor, while tirzepatide engages both GIP and GLP-1 receptors. The added GIP activity may contribute effects on insulin sensitivity and on fat metabolism. Whether the dual action produces meaningful clinical advantages beyond differences in potency is still being examined.
The downstream consequences of GIP receptor activation are not fully characterised in humans. It is also unclear how much each receptor contributes to appetite reduction and to shifts in body composition. Published work describes associations and proposed pathways rather than settled causal chains.
Water promotes hydrolysis and deamidation, so removing it slows degradation during transport and storage. The dry solid is also less prone to microbial growth than a solution. Reconstitution is therefore performed close to the point of use.
